Direct evidence for the spatial correlation between individual particle traversals and localized CDKN1A response induced by high-LET radiation

چکیده
In a recent study, we showed that heavy ion irradiation of human fibroblasts leads to an extremely localized nuclear response of the CDKN1A (p21) protein (1). The number of CDKN1A foci per cell nucleus after heavy-ion irradiation suggested a direct correlation with the number of particle traversals and therefore also a spatial correlation between foci and particle traversals. However, this conclusion was essentially based on statistical arguments, comparing the number of foci with the average number of particle hits which are expected at a given fluence and geometrical size of the nucleus. But using a method based on retrospective detection of traversals in nuclear track detection material, we were now also able to directly assign the individual foci to the sites of individual particle traversals (2,3). The procedure of fluorescence imaging and track image matching includes the following steps:

Direct evidence for the spatial correlation between individual particle traversals and localized CDKN1A (p21) response induced by high-LET radiation.
The spatial correlation between individual particle traversals and the nuclear CDKN1A (p21) response after high-LET irradiation of human fibroblasts was investigated. The experiments were based on a technique for the retrospective detection of particle traversals by means of nuclear track detectors, which were used as the cell substratum. This technique requires the precise repositioning of a s...

In order to assess the biological endpoints of late radiation effects like genomic instability and transformation it is crucial to understand the signal transduction mechanisms that connect initial DNA lesions with the overall DNA damage response. At the molecular level, high ionization densities result in an increased complexity of DNA lesions leading to an impaired repair ability and extreme ...

Background: The fatal effect of ionizing radiation on cells depends on Linear Energy Transfer (LET) level. The distribution of ionizing radiation is sparse and homogeneous for low LET radiations such as X or γ, but it is dense and concentrated for high LET radiation such as heavy-ions radiation. Material and Methods: Chinese hamster ovary cells (CHO-K1) were exposed to 4 Gy Fe-ion 2000 keV/...

Introduction: Microdosimetry is a fundamental method that studies the nature of energy transfer in micron volumes in the particular biological cells. In a biological target, the amount of ionization does not indicate the magnitude of biological radiation-induced damage. However, the severity of biological harm depends strongly on the amount of the linear energy transfer along t...
